Don't hype this issue...let go of the person. He has decided to go and is just looking for meagre issues...
Invest your time and effort to look for a suitable replacement.

From all your statements it is clear that this person wants to quit his job and is finding some junk reasons to justify his decision. I don't expect such a behaviour from a person who has worked for such a long time in a company, who don't even understand the basic policies.
I would suggest you to talk to his Wife regarding the issue, She may be understanding the situation. If this too is not working, then only choice is go ahead and say Good Bye to him.

I fail to understand how can such an irresponsible & immature man reach such a high level in your company.. When a person is at a Director level, it is expected from him to undersatnd not only his field of epertese, but also to have some knowledge of how other department work and what is their importance. You can't expect a sr. level person to tell you that HR is a waste of time and money and they do not do things properly. Didn't he understand the complications of this relationship when he got married. I think if he wants to leave, let him go his way.
Plus it is also advisable for couples to not work in the same company. It is neither good for the employees, nor for the employer.
If you still think, that the director is indispensable for the company, try to convince him, and shift his wife to some unassociated department. That might solve the problem of his feeling embarassed.

Please understand that "Nobody is indispensible for any company". May be this Director is very efficient at his work but I bet he is not a professional. I agree with few of the comments that may be he got another oppurtunity in hand & that could be the reason he is creating this mess.
Suggest that you allow him to leave after finding a replacement.

According to me if Director of your company wants to leave for this reason you should not stop him. Coz he can end up creating negativity in your office if he stays for atleast this what I percieve from his attitude. Reason being organisational culture always flow from top management & he being a director needs to understand & respect that. He needs to know that if your management would have celebrated director's wife birthday (entry level employee) in a same way than it would have caused high level of dissatisfaction among others employee who are giving same amount of effort . Thats how i see this organisation.
Well if you see it from my angle it would be better director leaves rite now.Else he could create problem later on for sure

I wud suggest - Make him understand the situation & ( His position & her position).
If he is understanding fine otherwise ask his wife to quit the job. ( This should be done thrugh him)
He is being the director, his wife (entry level) need not work for their living...
Let her be at home & let him celebrate her birthday in a ***** hotel.....
Otheriwise let him quit. if he does then we cannot find a fool in the world than him...
